PureCube Co-NTA Agarose XL
The term Co-NTA refers to a copper Ion that has been coupled Nitrilotriacetic acid. Co-NTA can then be coupled to agarose or magnetic beads for IMAC (Immobilized Metal Chelate Affinity Chromatography). This is a purification method to obtain functional His-tagged protein. Cube Biotech offers multiple different Co-NTA based products for protein purification. PureCube XL Co-NTA agarose consist of agarose beads with a diameter of around 400 µm. They are used for extreme cases of His tag protein purifications. These extremes cases are defined by very low flow rates. XL beads can be used for FPLC assays to overcome this issue. For Co-NTA based His Tag purification we recommend our PureCube Co-NTA resin with 40 µm diameter and our Purecube 100 Co-NTA agarose resin beads.
1. Protein Binding Capacity
The protein binding capacity is at least 5 mg/mL resin, as determined by purification of 6xHis-tagged GFP protein from E.coli cleared lysates, and quantified via spectrophotometry.
2. Compatibility
PureCube Co-NTA Agarose XL is very stable and can resist the following conditions in most situations: pH 2-14, 100% methanol, 100% ethanol, 8 M urea, 6 M guanidinium hydrochloride, 30% (v/v) acetonitrile.
